I love being on my own inside and outside of Amsterdam. I visit obscure coffeeshops on foot. I take the metro to the edge of Amstelveen and get high while admiring the cows. I take the train (10 minutes) to Zaandam and get high on the bike paths. I smoke on the hotel balcony in the evening and watch Dutch television.
It's precious. It's time I actually enjoy spending with myself.

I've gone on my own once and with mates/gf the other 5/6 times
Pros
You can do exactly what you want when you want
You will probably do more activities rather than sit around in coffee shops all day
You are more likely to chat to randoms
Cons
Slightly depressing smoking on your own in a coffee shop, most people are in groups
Holidays/trips are best enjoyed when you are with those you care for IMO

I spent the first 36 hrs of my last trip in Amsterdam alone... while it was fun, after a bit I missed being able to talk to a friend... no offence to my AMS friends... but I think you understand, it's a little bit of "stir-crazy" for me after a day.
everyone is different tho...

My tip for a solo trip would be to pay a little extra and get a nice big room with a great view of the hustle and bustle. This way you can hang out comfortably and relax during downtime. When there is down time when you're home you know what to do, but on a solo trip you wonder what the hell to do. A nice room helps with that as a nice refuge.

I'm considering travelling over in 2 weeks time... The only thing that I have been debating in my head is whether or not to stay for one night or two? I've never travelled alone but I do quite enjoy smoking alone and doing things myself.. do people reckon one night would be enough to satisfy my cravings??

If you're not traveling with anyone you can feel free to spoil yourself by doing anything you please, at any time...
Visit a spa...

Check out a movie at the impossibly ornate Tuschinki Theater...

I ate a brownie before going to see Jim Carrey's Liar Liar there (yeah, it was awhile ago) and the art deco exuberance, along with Jim Carrey's facial contortions, really tripped me out.
Learn some Dutch before going and practice while you're there. Harder to do with friends, unless they're Dutch, of course.
Google Translate...
http://translate.google.com/
They've got a smart phone app too.
Save bookmarks in your browser of interesting places and access them while there. I also take advantage of Google Earth by making and saving a collection of placemarks, and geo-locating them on my mobile device while there. That's what we did when we walked to the Cannabis Cup Expo at the Borchland.
If you haven't made a reservation yet, try a stay with someone who's renting a space. They'll be able to give you a perspective only a local has...

I'll be spending my 20 nights in May/June doing just that, at two different places (two different perspectives).
The Nes-Cafe, among others, is always a great place to go and meet new folks.
Put on your adventure hat and go for it!
Cheers
